I sent in an application for purchase of tusmitate nisa eMAXIS Slim 米国株式(S&P500) on Dec 14, 2021.
However, the status of the application is still at 積立状況 設定中
Is it supposed to take so long to process the application?

Yes, it just takes a while to get the NISA set up.

Mine took around 2 months, iirc.
